  Maedler et al. (2004) observed decreased expression of IL1RN in human pancreatic beta cells of patients with type 2 diabetes (see 125853) compared with nondiabetic individuals. In vitro, chronic exposure of human islets to leptin (164160) decreased beta cell production of IL1RN and induced IL1-beta release from the islet preparation, leading to impaired beta cell function, caspase-3 (600636) activation, and apoptosis. Exogenous addition of IL1RN protected cultured human islets from the deleterious effects of leptin; antagonizing IL1RN by introduction of small interfering RNA (siRNA) into human islets led to caspase-3 activation, DNA fragmentation, and impaired beta cell function.
